Title

FORMULATION AND DEVELOPMENT OF EMTRICITABINE PELLETS BY EXTRUSION SPHERONIZATION METHOD

Abstract

Emtricitabine (commonly called FTC, systematic name 2',3'-dideoxy-5-fluoro-3'-thiacytidine, with brand name Emtriva (formerly Coviracil), may be a nucleoside reverse-transcriptase inhibitor (NRTI) for the prevention and treatment of HIV infection in adults and youngsters. A fixed-dose triple combination of emtricitabine, tenofovir and efavirenz (Sustiva, marketed by Bristol-Myers Squibb) was approved by the U.S. Food and Drug Administration (FDA) on July 12, 2006 under the name Atripla. Emtricitabine is indicated together with other antiretroviral agents for the prevention and treatment of HIV infection in adults. Emtricitabine is commercially available and is approved by the FDA for treatment of HIV infection. Pellets were prepared by extrusion spheronization technique using other ingredients like Avicel PH101 as filler, lactose monohydrate as pore forming agent and ethyl cellulose solution as binder. Box Behnken design was used for optimization study. Pellets showed excellent flow properties and good sphericity.
