Title

NEUROPROTECTIVE EFFECT OF MEFENAMIC ACID IN EXPERIMENTAL MODEL OF ALZHEIMER’S DISEASE

Abstract

Neuroinflammation has gained substantial attention for its involvement in the progression of Alzheimer’s disease (AD). Recently platelets have been reported to stimulate the progression of AD by contributing to 90% of circulatory Aβ. Therefore, we hypothesized to use mefenamic acid, an anti-platelet and anti-inflammatory drug for the treatment of AD. We investigated the effects of mefenamic acid on STZ induced cognitive decline and neuroinflammation. For in-vivo studies, intra-cerebroventricular streptozotocin (ICV-STZ) (3mg/Kg) induced Wistar rat models of AD were incorporated in the study. Rats were divided into 3 groups of 5 animals each, including sham, STZ and mefenamic acid group. For in-vitro studies, BV2 cells were incorporated and subjected to MTT and RT-PCR. Behavioural studies revealed that mefenamic acid ameliorates the cognitive deficit associated with STZ. Anti-platelet effect of mefenamic acid was also determined by using tail vein bleeding assay. Further, in-vitro studies on BV2 cells suggested that mefenamic acid potentially regulated the expression of pro- and anti-inflammatory cytokines. Therefore, findings of this study suggest that mefenamic acid has a potent neuroprotective effect by virtue of its anti-platelet and anti-inflammatory activity.
