Title

Characterization of Carrageenan Extracted from Kappaphycus alvarezii and its Potential Application in Plant Culture Medium

Abstract

Kappaphycus alvarezii (Doty) is a marine macro algae has economic value, which is considered as source of carrageenan. The present study thus implies a standardization of methods for preparation of carrageenan from red seaweed Kappaphycus alvarezii. Based on experimental screening hot alkali treatments influence the yield, and extracted samples were subjected to structural analysis such as scanning electron microscopy to identify the morphological features, FT-IR and H-NMR for prediction of compound based on its functional groups as well as shift of peak compared with commercial standard. The results indicated that different extraction process determine the production of carrageenan from Kappaphycus alvarezii. On comparison of its significant role in plant tissue culture medium were analysed with agar at various time factors in order to confirm its potential in the replacement of agar from MS medium. Carrageenan is gelling nature useful in conservation of solution consistency as solidifying agent in tissue culture medium. Experimental results revealed that carrageenan can be a potential compound act as solidifying agent as well as proliferation of plant tissues through induction of callus, proliferation of shoot and root of explant in MS culture medium addition of carrageenan instead of agar. Present finding suggested that the utility of carrageenan from Kappaphycus alvarezii proved to be a promising novel compound in chemical production industry particularly in the application of plant tissue culture medium composition.
