Somaclonal variation may appear in regenerants due to various factors associated with the in vitro culture conditions. Checking genetic stability of the in vitro clones is obligatory if we are to propagate true- to- type quality planting materials. Different molecular markers have been employed for clonal fidelity testing of in vitro propagated plants. These are DNA markers which are PCR based such random amplified polymorphic DNA (RAPD), amplified fragment length polymorphism (AFLP), inter- simple sequence repeats (ISSR), simple sequence repeat (SSR) and start codon targeted (SCoT) among others. The present mini review focuses on the description of varied molecular markers, their methodology, strengths and limitations as well as their applications in genetic homogeneity testing of in vitro regenerants belonging to different plant species.
